Generation, differentiation, and functional features of organ-specific macrophages and the impact of macrophage ontogeny on interaction with metastasizing tumour cells

Objectives:

  1. Establish standardized protocols for iPSC-derived organ-specific macrophage generation and functionality testing
  2. Develop strategies for populating OoCs with organ-specific macrophages and testing their functional capacities
  3. Determine organ-specific targets of macrophage-tumour cell interactions

Expected Results: Human organ-specific macrophages (Kupffer cells, microglia, osteoclasts) will be generated from human iPSCs via differentiation protocols using cytokine/growth factor cocktails and/or infiltration of pre-cursors into organotypic environments. Assays to test general and organ-specific functional responses will be determined. The integration of organ-specific macrophages into OoC models will be established, and the impact of infiltrating tumour cells on their functional parameters in OoC models will be determined, including identification of molecular markers.
