Identification of macrophage function and induced by the first interaction with invading neuroblastoma (NB) cells in the brain and liver OoC model
Objectives:
- Establish standardized human brain OoC integrated with organ-resident (microglia) and blood monocyte-derived macrophage population
- Identify macrophage and matrix changes induced upon interaction with NB cells in brain and liver OoC model
- Select and test potential macrophage-targeted compounds to interfere NB cell survival and invasion
Expected Results: iPSC-neural stem cells (iPNSCs),-astrocytes (iPSCAs), and -microglia (iPMic)/ blood monocyte-derived macrophages will be pneumatic droplet-dispensed using RegenHU R-Gen 200 3D printer in 96 well plates which will subsequently form nascent spheroids. These mini-brain spheroids and liver model from DC1 will be transferred into the optimised and controlled (MIVO) OoC system to validate the very early biomarkers of macrophage changes induced by NB cell interaction and impacting macrophage function and matrix composition (WP2).
