Development and optimization of a multicompartmental OoC with macrophages neuroblastoma cancer (NB) cells in circulation around brain and liver target tissues

Objectives:

  1. To develop an multicompartmental OoC integrated with (age-relevant) organ-resident and blood monocyte-derived macrophage population;
  2. To recapitulate the cancer cells infiltration and survival in a physiological capillary flow
  3. Test tissue-specific hydrogel options
  4. Select and test potential macrophage-targeted compounds in the OoC platform

Expected Results: We will set up and validate the 3D dynamic OoC MIVO culture of human brain and liver enriched with the immune component. Different fluid flow dynamic conditions will be set up (e.g. fresh immune cells media circulation or close loop circuit) with different matrixes resembling the tumour microenvironment. We will evaluate immune cell infiltration and activation, by measuring specific biomarkers. Subsequently, we will test tumour cell invasion using microscopic and cytofluorimetric analysis and test macrophage-targeted effects.
