Identification, characterisation and manipulation of glycosaminoglycans (GAGs) in the ageing matrix
Objectives:
- Optimise in-situ GAG analysis using ORBI-SIMS
- Work with partners to identify GAG changes induced by the macrophage-tumour cell interaction
- Test impact of modulators of GAG synthesis in in vitro models
Expected Results: GAGs have multiple roles in controlling cell behaviour in healthy tissue which can be subverted in disease. For the first time, we are able to use MS-imaging in an unbiased way to detect changes in GAG levels and composition using MS-imaging in an unbiased way to detect changes in GAG levels and composition on tissue ageing and invaded and non-invaded tissues, which can then be interpreted to build testable hypotheses for potential targets in the GAG biosynthetic pathway.
